Development of a novel self-amplifying circular RNA vector for durable transgene expression

The potential use of RNA-based therapeutics is rapidly increasing. However, lipid nanoparticle (LNP)-delivered mRNAs have a relatively short half- life in vivo and transgene expression beyond 3-5 days is minimal. Therefore, novel RNA modalities that can sustain stable transgene expression for extended periods would open a wealth of therapeutic options. This study will explore the development of a circular single-stranded RNA delivered in a simple LNP-format to achieve long-term expression of a transgene.
